Roles of glutathione and L-cysteine in the biomimetic green synthesis of CdSe quantum dots

摘要: Biological synthesis of quantum dots (QDs) as an environmental-friendly and facile preparation method has attracted increasing interests. However, it is difficult to distinguish the roles of bio-thiols in QDs synthesis process because of the complex nature in organisms. In this work, the CdSe QDs synthesis conditions in organisms were reconstructed by using a simplified in vitro approach to uncover the roles of two small bio-thiols in the QDs formation. CdSe QDs were synthesized with glutathione (GSH) and L-cysteine (Cys) respectively. Compared with Cys at the same molar concentration, the CdSe QDs synthesized by GSH had a larger and broader particle size distribution with improved optical properties and crystal structure. Furthermore, quantum chemical calculations indicate that the stronger Cd2+ binding capacity of GSH contributed a lot to the CdSe QDs formation despite of the greater capability Cys for selenite reduction. This work clearly demonstrates the different roles of small thiols in the Cd2+ stabilization in the environment and biomimetic QDs synthesis process.

Effects of Decoction on plasma proteome in cirrhosis: preliminary experimental study with rats

摘要: The aim of this paper is to study the effects of Decoction on the plasma proteome in cirrhotic rats. Twenty-six male Sprague-Dawley (SD) rats were randomly divided into three groups: cirrhotic model group ( = 10), treated with CCl (CCl/olive oil: v/v = 1:1); Decoction intervention group ( = 10), treated with CCl+ Decoction; and normal control group ( = 6), treated with olive oil only. After 8 weeks, blood samples were collected from the inferior vena cava to undergo bi-dimensional electrophoresis (2DE) and analysis by PDQuest 7.3 software. Differential protein spots were cut, enzyme hydrolysis was conducted, and peptide fragments extracted from the mixture underwent mass spectrometry (MS) with MALDI-TOF-TOF-MS. The liver fibrogenesis was assessed using a digital image analysis instrument of Masson’s trichrome stained sections. The fibrosis area of the Decoction was (8.9 ± 3.7)%, significantly smaller than that of the cirrhotic model group [(12.4 ± 4.7)%, < 0.05]. Ten markedly changed protein spots were identified by MALDI-TOF-TOF-MS. Eight of the 10 proteins, including plasma glutathione peroxidase, plasma glutathione peroxidase precursor, prealbumin, haptoglobin, apolipoprotein A-IV precursor, complement C4, inter-alpha-inhibitor H4 heavy chain, and serine/threonine-protein kinase microtubule-affinity regulating kinase 1 (MARK1) were expressed very lowly in the cirrhotic model group while they were expressed highly in the Decoction group. The expression of liver regeneration-related protein LRRG03 and vimentin increased in the cirrhotic model group, and reduced in the Decoction group. Some proteins related to oxidative stress, cell proliferation and transformation have changed in the plasma of cirrhosis induced by CCl. Decoction promotes protein synthesis and plays an anti-fibrotic role by anti-oxidation and accommodation of cell proliferation and transformation.

Analysis of two new degradation products of arsenic triglutathione in aqueous solution

摘要: Inorganic arsenicals, including arsenite (As ) and arsenate (As ), are well-known human carcinogens. Recently, studies have indicated that arsenic triglutathione (As(GS) ) is unstable in an aqueous solution. The present study was designed to evaluate the degradation mechanism of As(GS) in an aqueous solution using high-performance liquid chromatography-electrospray ionisation mass spectrometry (HPLC-ESI-MS). Based on the fragments obtained from MS and MS , we identified two new compounds: one was an isomer of glutathione (GSH), and the other was a product from the cleavage of the glutamyl of oxidised glutathione (GSSG). The isomerization of GSH resulted in the loss of its function such as detoxification of many reactive metabolites. The formation of the two new compounds affected the ratio of GSH/GSSG, and thus may affect the antioxidant and detoxification of GSH/GSSG in mammalian cells.

Mechanism of hepatocellular damage in rat caused by low serum selenium

摘要: The aim of this paper is to investigate the mechanism of hepatocellular damage in rats caused by low serum selenium. Thirty six rats were randomly divided into 2 groups: group A (fed with low-selenium diet from the Keshan Disease area with the content of selenium being 0.017 mg/kg); group B [fed with selenium-supplemented diet and 0.3 mg/L selenium (NaSeO) was added to the drinking water]. Both were respectively fed for 12 weeks. At the end of the 12 week, the levels of serum selenium, glutathione peroxidase (GPX) and malondialdehyde (MDA) in hepatic tissue were measured; the hepatocellular ultrastructure and apoptosis were observed as well. The levels of serum selenium and GPX in group A were markedly lower than those in group B. MDA level in group A was significantly higher than that in group B. Under the electron microscope (EM), the mitochondria were remarkably changed in group A. The rate of liver cell apoptosis appeared much higher in group A as well. It indicated that the damage caused by selenium deficiency was through the process of oxidation. Selenium deficiency led to apoptosis of hepatocytes where oxidative damage to mitochondria might be the cause.
